Title: Monocytes and apoptosis of vascular endothelial cells in vitro

Abstract:

The aim of my research was to reveal the involvment of monocytes in vascular endothelial cells apoptosis in vitro. The purpose of the study had to be achieved by: assesment of the mRNA and protein expression for proapoptotic PUMA and heat shock proteins HSP70 and HSP27 in HUVEC cells; demonstration of protein AIF nuclear translocation; assesment of DNA breaks; demonstration of executioner caspases products (PARP) in HUVEC. Additionally, levels of PUMA, HSP70, HSP27 had to be assesed in HUVEC exposed to proinflammatory cytokines IL-1[beta], IL-6, MCP-1 and TNF-[alfa]. HUVEC cells were used as human vascular endothelial cells model. Monocytes model were both the MonoMac6 and monocytes isolated from the peripheral blood. Two types of mixed cultures were used: direct and insert. After 24 h and 48 h of coculture, monocytes were discarded and HUVEC cells were subjected to RNA or protein (cytoplasmatic and nuclear) extraction or the procedure of detection of apoptotic cells. The main achievement of this work is that monocytes do not induce the HUVEC apoptosis despite the fact, they up-regulate PUMA level. The presented results indicate a crucial cytoprotective role of HSP27, its form p-HSP27 and HSP70 action, because their levels are significantly increased in HUVEC exposed to monocytes. A similar observation made in HUVEC treated with cytokines makes possible the idea, that in vascular endothelial cells a complex mechanism appears. Probably apoptotic cascade and stress protein system (responsible for cellular homeostasis) are activated simultaneously and due to the action of cytoprotective proteins, HUVEC cells survive.
