Title: Influence of the C-MET, HGF/SF on invasiveness and prognosis in rhabdomyosarcoma

Abstract:

C-MET-HGF/SF axis plays an important role in the pathogenesis and progression of many cancers, including sarcomas. That is why the influence of C-MET-HGF/SF axis on progression of the most common sarcoma in childhood-rhabdomyosarcoma (RMS) was taken under consideration. In this material expression of the C-MET receptor, HGF/SF ligand was examined, as well as HGF/SF influence on proliferation/viability and migration of the established RMS cell lines. The presence of the potential activating mutations in the kinase domain was assessed by sequencing. The statistical relation between the expression of the C-MET and survival was performed. The expression of the C-MET receptor in the RMS cells was observed. No expression of the HGF/SF was found in the material, so there is no activating loop in the progression of the tumor. There was no mutation found in the kinase domain of the C-MET receptor. HGF/SF increased the motility and direct migration of the RMS cells, as well as the proliferation and viability. The presence of the translocation PAX3-FKHR was observed in the alveolar subtype of RMS (ARMS).
