Title: The influence of hematopoietic cell preservation conditions on their hematological potential in experimental model


A lot of elements have influence on the way of cells preparation for transplantation, including conditions in which cells are stored. The aim of this work was to study the influence of anticoagulants such as EDTA, CPDA-1 and heparin, temperature 4oC and 37oC as well as timing of cells preservation: 0, 1, 6, 12, 24 h on their response to stromal derived factor-1 and also ability of progenitor to form granulocytes-monocytes colonies. Study material was bone marrow received from mice type Balb/c. The analysis was executed by following methods: chemotaxis, flow cytometry, clonogenicity tests. Experiments conducted showed that: - ability of hematopoietic cells to migrate and to form granulocytes-monocytes colonies (CFU-GM) in vitro depends on type of anticoagulant, temperature and storage time, - heparin is best suited anticoagulant for bone marrow cells preservation, - bone marrow cells preserved in 37oC demonstrated better migration response and proliferative potential towards granulocyte-monocyte line, - higher migration and number of CFU-GM colonies in 6-hour-storage were noticed, - number of migrated cells and colonies is changing periodically during 24-hour bone marrow storage, - the best growth of CFU-GM in group of cells incubated with heparin over 6 h in temperature 37oC was proved. Obtained colonies were the most numerous and the biggest from among tested groups. This result can indicate on a better hematopoietic recovery concerning granulocyte-monocyte line.
