Title: Expression of vascular endothelial growth factors: VEGF-A, VEGF-C, VEGF-D and their receptors: VEGFR-1, VEGFR-2 , VEGFR-3 in cutaneous malignant melanoma
Abstract:
Aims: To establish the correlation between expression of vascular endothelial factors: VEGF-A, VEGF-C, VEGF-D, their receptors VEGFR-1, VEGFR-2 , VEGFR-3 as well as blood and lymphatic microvessel density and morphological prognostic factors for melanoma. Materials and methods; 423 cases of primary malignant melanoma were examined. Microvessels were stained with CD31, CD105 and D2-40 antibodies. Blood and lymphatic microvessels were counted both, peri- and intratumorally. For CD31 and D2-40 positive vessels Chalkley count technique also was applied. Immunohistochemical expression of VEGF-A, VEGF-C, VEGF-D and receptors VEGFR--1,-2,-3 in tumor cells, as well as blood and lymphatic microvessel density were correlated with morphological prognostic factors and patients survival. Results: There was no positive correlation between tumor thickness, ulceration and VEGFs and their receptors expression. Expression of growth factors and their receptors showed no prognostic value in univariate analysis. Intra and peritumoral blood microvessel density as well as Chalkley count for intratumoral blood microvessels were correlated with tumor thickness and presence of ulceration (p<0.01). In univariate analysis higher Chalkley count and higher blood microvessel density was associated with shorter disease free (p<0.014) as well as overall survival (p<0.003) Conclusion: In malignant melanoma, the ; complex assessment of the blood and lymphatic microvessel density seems to be more useful in terms of prognostication than evaluation of expression of selected vascular endothelial growth factors and their receptors.
