Title: The influence of the selected fatty acids on expression gene of aromatase in progenital cells of human adipose tissue

Abstract:

Since the end of the last century the adipose tissue has been considered as an organ of endocrine secretion. A number of key substances – adipokins are produced by the cells of the adipose tissue – adipocites, and the stromal vascular fraction of adipose tissue. They participate in integration and co-operation of various tissues and supervise metabolic processes. The most important adipokins and cytokins are: leptin, adiponectin, adipepsin, adipsin, resistin, interleukin 6 (IL-6), inhibitor of plasminogen type 1 (PAI-1) activator, TNF, angiotensinogen, as well as enzymes, such as aromatase. Aromatase (P-450 aromatase) is the main enzyme of oestrogen synthesis in all vertebrates. Oestrogens synthesized in the presence of aromatase regulate the functions connected with sexual features and show a protective activity for the cardiovascular system. They also have pro-carcinogenic activity in case of breast and endometrial cancers. There is a significant difference in aromatase expression in women depending on their age. In pre-menopausal period the sites of aromatase expression are ovaries, whereas in postmenopausal period – the visceral and subcutaneous adipose tissue. Taking into consideration previously known the role of this enzyme in oestrogen production in females, the aim of this work was investigation of the difference in aromatase expression in the visceral and subcutaneous ; adipose tissue in women in pre- and post-meopausal period. Being aware of the protective influence of diet in prevention of the diseases of the circulatory system, we also studied the influence of dietary fatty acids (in non-toxic concentration 10-30μM) on aromatase expression. These fatty acids are arachidonic acid (AA) eicosapentaenoic acid (EPA), olein acid (OA), palmitinic acid (PA). The effect was important for the stromal vascular fraction (SVF) of the adipose tissue which is considered to contain the most metabolically active cells. The aim of the work was to indicate the difference in aromatase expression in the subcutaneous and visceral adipose tissue in women in pre- and peri-menopausal period. Moreover, in in vitro investigations, the influence of saturated mono- and poly-unsaturated fatty acids on aromatase expression in SVF cells isolated from the human subcutaneous and visceral adipose tissue was studied. Assessing the results of in vitro investigations, it has been found that there is a positive correlation between the age of the patients and their BMI value, and hence aromatase expression is dependent on the age and the abdominal obesity. In in vitro investigations it has been shown that saturated fatty acids induce aromatase expression in progenitor cells, especially those which differentiate to adipocites. In SVF cells directed towards endothelial cells, the ; adverse reaction has been observed.
