Title: The concentration of osteopontin in the patients' blood serumwith clinically and histologically confirmed endometriosis

Abstract:

Endometriosis is a disease that affects about 2-10% of women in premenopausal age. Her main symptoms are infertility and chronic pain. To this day, its pathogenesis remains unknown. There is also no marker that would facilitate the detection of the disease. Osteopontin is a glycoprotein that plays a role in processes such as angiogenesis, apoptosis or the development of inflammation. In the study, we examined the correlation between serum osteopontin concentration in patients with endometriosis and osteopontin gene expression in endometrial implants taken during laparoscopy. We also assumed that osteopontin can be a marker of endometriosis, the concentration of which can be examined in the blood. The collected implants were analyzed using the microarray technique. Control was a healthy peritoneum taken during the same procedure. Microarray analysis showed increased expression of the osteopontin gene in the diseased tissue. The level of osteopontin mRNA expression was estimated by real-time PCR. The concentration of osteopontin in the blood of the patients was measured by the enzyme immunoassay ELISA.The results of the study showed increased expression of the osteopontin gene in the presence of endometriosis, confirmed in the tissue by RT-PCR expression. The concentration of osteopontin in the serum of healthy patients and patients also showed differences, it was higher in women p ; atients, which may postulate its role as a biomarker of endometriosis.
