Title: Hydroxyapatite/polylactide composite material in the healing model of bone defects in rabbits

Abstrakt:

The aim of the present study was to:1. identify the optimal proportions – for the purposes of bone reconstruction – between ceramic and polylactide elements in osseosubstitute material, prepared in collaboration with the Crystal Chemistry of Drugs Team of the Faculty of Chemistry of the Jagiellonian University 2. assess the cytotoxic activity of the material 3. describe a model for the healing of bone defects in rabbits using a ceramic-polylactide composite4. provide a histological assessment of the effects of a ceramic-polylactide composite on the healing of bone defects in rabbits. The testing of the ceramic-polylactide material comprised both a laboratory and an experiment part.1. The method employed in the study proved a useful tool for achieving the study objectives.2. The results of the cytotoxicity tests indicated that the composite material did not have any toxic effect on the surrounding tissue.3. Compared with control group K the bone defects in groups A and B, which were filled with a ceramic-polylactide material, healed with less inflammatory infiltration, faster regression, no resorption or bone necrosis, which ensured better regeneration of bone tissue.4. A statistical analysis of the study results showed increased resorption of the composite in group B, which might have been due to the higher polylactide content. Simultaneously, bone defects filled with ceramic-p ; olylactide composite in proportions of 80/20 and 61/39 healed at comparable rates.5. The use of ceramic-polylactide material induces mesenchymal cell differentiation into osteoblasts, which in turn initiates the processes of osteogenesis. This mechanism has not yet been fully explained and should be the subject of further research.
