Title: Microparticle as a new source of signals for stem cell "homing" and neovascularisation

Abstract:

Recent works demonstrated that microparticles formed by activated platelets (PMP) may serve as a new source of signals "guiding" progenitor and neoplastic cells in vivo. Moreover PMP were shown to induce angiogenesis in vitro and in vivo. This properties of microparticles permit to hypothesize that PMP can regulate the biology of endothelial progenitor cells and play role in neovasculogenesis and angiogenesis. Obtained data demonstrated that PMP can regulate homing of endothelial progenitor cells by inducing migration and adhesion of this cells, as well as by promoting invasion of such cells to the places of neovascularisation by stimulating expression of MMP-9 PMP regulated progenitor cells biology by transfer of adhesion molecules from PMP to membrane of target cells. It was shown for the first time, that PMP can also increase the expression of VCAM-1 on endothelial cells surface. In this work it was demonstrated that PMP induced proliferation and differentiation of both endothelial progenitor cells and mature endothelial cells (HUVEC). PMP exe11ed their effect on cells via activation of Akt and p42/44 MAPK This study also revealed the new role of CD36 receptor presented on PMP surface in regulation of trombospondin-1 (TSP-1) dependent cell apoptosis. Based on observations demonstrated in this work, it can be suggested that PMP presented in a high concen ; tration at the sites of wound healing and inflammation are involved not only in process of coagulation and inflammatory response, but could also participate in vessels regeneration. Therefore PMP could be potentially used as a new factor for stimulation of tissue renewal.
