Title: Analysis of genes SERCA2A and angotensiongen and growth factors in patients with left ventricular hypertrophy in curse of essential hypertension


The study detected M235T (c.704 C>T) polymorphism of AGT gene, searched for mutations of ATP2A2 and for frequency of these polymorphisms in hypertensive patients in comparison to control group. The blood level of TGFB1, bFGF, IGF-1 was measured and compared between these groups. The 157 people aged 52.33±1,22 year took part in the study. The echo examination and ABPM were performed. The M235T polymorphism of AGT was analysed with real-time PCR and TaqMan sonds. The polymorphisms of ATP2A2 were detected using dHPLC and sequencing analyse. ELISA served for detection of TGFB1, bFGF and IGF-1. We did not find any correlation between the level of TGFB1, bFGF, IGF-1 with blood pressure values and LVH development. The frequency of homozygotes CC of M235T polymorphism of AGT was the highest in group with LVH. The allel T carriers had higher value of blood pressure measured in cabinet; and systolic blood pressure in ABPM. The results seem to confirm promoting influence of M235T polymorphism on predisposition to development of hypertension. The new polymorphism A724A (c.2171G>A) in ATP2A2 exon 15 was detected. The presence of genotype GA was significantly higher in control group in comparison with other groups. GA carriers demonstrated lower level of left ventricular parameters and blood pressure value measured in cabinet and in ABPM. The c.2171G>A polymorphism seems to play the protect ; ive role against hypertension and LVH. The results may be useful in detection of panel of candidate genes that increase risk of hypertension and LVH, useful for creation of genetic tests.
