Title: Analytical assessment of azole antifungal drugs

Abstract:

The subject of this dissertation is analytical assessment of selected azole antifungal substances; bifonazole, econazole, fluconazole, itraconazole, ketoconazole, clotrimazole and miconazole. Elaboration of new methods of qualitative identification, quantitative determination, separation and also elements of purity assessment was done. For above mentioned purposes three analytical techniques were used; UV-Vis spectrophotometry including derivative spectrophotometry, thin-layer chromatography (TLC) with densitometric detection and capillary gas chromatography with flame-ionisation detection (CGC-FID). Methods presented in this thesis were validated and their usefulness in routine pharmaceutical analysis is proven. Many innovative procedures were described. Spectrophotometric procedure for itraconazole is prepared for the first time. Bifonazole, econazole, ketoconazole, clotrimazole and miconazole were not determined by using CGC-FID till now. Researches with itraconazole on gas chromatography, presented in this dissertation, could be useful in additional qualitative analysis. Separation of all investigated azoles done using GC and separation for 4 azoles (bifonazole, fluconazole, itraconazole and ketoconazole) by TLC is also innovate. Elaborated methods are valuable alternative and supplementation of investigational possibilities in the group of azole antifungal drugs for the ro ; utine pharmaceutical analysis purposes. They are important for quality assurance of substances under consideration.
