Title: Influence of PPARgamma agonist on expression of adiponectin and its receptors during differentiation of preadipocytes, vessel endothelial cells as well as progenitor cells isolated from human adipose tissue

Abstract:

Adipose tissue is a highly active endocrine organ secreting a range of compounds with both local and distant actions. Adiponectin is one of the endogenous adipose-derived factors, named adipokines, which has been demonstrated to play an important role in the modulation of the liver glucose and lipid metabolism. Recently two types of adiponectin receptors have been found. In the contrary to leptin, the decreased plasma level of Adq is observed in patients with obesity and type 2 diabetes. Adiponectin exerts many anti-inflammatory and anti – atherogenic properties. Modulation of the adiponectin level in plasma by nutrients (fatty acids, glucose) and insulin-sensitizing drugs (PPAR agonists ie glitazones) may support the normal blood/vessel wall homeostasis. The stromal vascular fraction - SVF play an essential role in the adipose tissue mass since it consist of the adipocyte and endothelial progenitor cell fraction. In this project the influence of FFA and some PPAR agonists on expression of adiponectin and its receptors in the adipose tissue derived SVF was investigated. Isolation and culture of human stromal adipose tissue progenitor cells was done using modified Hauner method. After checking the cytotoxicity of examinated substances, the influence of FFA and PPAR agonists on proliferation, migration, tube formation as well as differentiation to adipocytes of investigated cells ; was studied. The relative expression of investigated genes was prepared using real-time PCR followed by Western Blot analysis of proteins level. Changes of adiponectin amount released to medium by differentiating SVF cells were measured using Adiponectin Elisa kit. During the differentiation process the expression of adiponectin and its receptors was differentialy modified by FFA and PPAR agonists, which was shown in real-time PCR as well as Western Blot. Some of used factors changed also amount of adiponectin released to medium by differentiating SVF cells. These results suggest an additional mechanism of beneficial effect of unsaturated fatty acids (especially EPA) as well as some PPAR agonists, follow by the influence on expression of adiponectin and its receptors, which is important for endothelial function as well as insulinsensitivity. Simultaneously the disadvantageous effect of saturated palmitic acid has been confirmed.
