Title: Quantitative determination of aminoglycoside antibiotics by derivative spectrophotometry

Abstract:

The PhD thesis presents reactions of aminoglycoside antibiotics with ortho-phthalic aldehyde, ninhydrin and fluorescein and their application for quantitative determination by derivative spectrophotometry. Modification conditions for analized compounds have been developed (temperature, time of reaction). The mechanism of reaction has been established taking into consideration reagents' structure and results of 1H NMR. Mole ratio of reagents has been determined. Absorption spectra were measured, analytical wavelength was chosen and the applicability of obtained compounds for spectrophotometric analysis was checked. In order to mcrease the accuracy of the determinations, absorption spectra were converted into derivative spectra for further analyses. The following derivatives were chosen: after reaction with ortho-phthalic aldehyde third-derivative spectrum, with ninhydrin first-derivative spectrum, and with fluorescein second-derivative spectrum. Precision of the method was good, RSD <lid not exceed 3.6%. Linearity range was determined for each examined antibiotic. The correlation of calibration curves was higher than 0,99. Accuracy was determined based on the recovery and was good for all antibiotics. 4 Calculated values of LOD and LOQ (· 10-) prove that the method is highly sensitive. The content of gentamicin, amikacin and neomycin has been determined in chosen pharmaceutica ; l preparations. The determined content was close to the content declared by manufacturer. The application of derivative spectra enabled to eliminate the influence of excipients present in pharmaceutical preparations, with spectra superimposing on spectrum of active substance.
