Title: Vagus nerve participation in different models of pain

Abstract:

Pain management is difficult and still not highly effective, for that reason alternative ways of pain soothing need to be explored. The vagus nerve can be a target for that therapy. The aim of investigation were to evaluate the participation of vagus nerve in experimental pain models like visceral pain (VP), neuropathy (CCI) and neurogenic inflammation (NI). We analysed modification of vagal nerve activity (electrical activity and c-fos expression) and behavioural changes in response to nociceptive stimuli. Influence of vagal component was identify by comparison to the appropriate groups with executed vagotomy. Influence of inflammatory component was regulated by pre-emptive blockade of pro-inflammatory cytokines via pentoxifylline (PTX). In VP model we observed the direct participation of the vagus nerve in mechanisms of visceral nociception and responsiveness with inflammatory component in the development of peripheral and central sensitization. The vagus nerve also plays a role in sensitization processes in the development of chronic pain complexes (CCI) by cooperation with the immune system on the central level. In the NI model the vagus nerve doesn't participate in mediation of mechanical allodynia, and even more can increase the development of the thermal hyperalgesia. These results can prove central blockade of vagal centres to facilitate the nociceptive information tran ; smitted by spinal pathways. Our results establish the participation of vagal component in nociception what can constitute the base for using this pathway in acute and chronic pain syndrome treatment.
