Abstrakt:

In our investigations, the topical use of antidepressants (amitriptyline, doxepine, venlafaxine) on neuropathic pain development was examined. Neuropathic pain, the result of neuropathy, was evoked by constriction chronic injury of the sciatic nerve in rats. The results of locally administered antidepressants on pain behavior were assessed. Antidepressants used in the study were administered into the ventral part of paw affected with neuropathy. For the study of pain behavior we used von Frey’s test for allodynia and paw withdrawal test for hyperalgesia. Analysis of interactions between topically administered antidepressants and morphine and influence of serotoninergic, adrenergic and opioid systems on effects of antidepressants were examined. All three studied drugs reduce development of allodynia, but only tricyclic antidepressants reduce hyperalgesia. We found that inactivation of opioid, adrenergic and serotoninergic systems abolishes the inhibitory actions of the studied drugs on development of allodynia or hyperalgesia. The analgesic effects of morphine were potentiated by concomitant administration of amitryptyline and attenuated by coadministration of doxepine or venlafaxine.
