Title: Oral microbiome of patients after hospitalization due to COVID-19 in a metagenomic seąuencing (mNGS) study along with standardization of the method of obtaining biological material

Abstract:

The oral microbiota varies depending on specific areas, such as the tongue, cheeks, palate, tonsils, teeth, and periodontal pockets. The lack of standardization in the selection of sampling sites and conditions poses a challenge in comparing the results of different studies, making it difficult to analyze microorganisms and their typing in the context of potential disease biomarkers. Previous research indicates that poor oral hygiene and severe periodontal disease may worsen the course of various infections, including SARS-CoV-2. The main aim of the study was a qualitative and semi- quantitative assessment of the composition of the oral bacteriobiome using next-generation sequencing in groups of patients recovered from C0VID-19 who received an antibiotic during hospitalization (Group I) and those who did not receive an antibiotic during hospitalization (Group II), compared to a control group (study participants who did not have COVID-19). An assessment of oral health status using selected dental indicators was also performed on all study participants. As a result of the study, protocols were developed describing the conditions for preparing patients for collecting biological materials, a diagram of the dental examination with details of the method and sequence of collecting materials, as well as the conditions of transport and storage of the collected sampłes until microbiologi ; cal tests are performed. It was observed that COVID-19 patients had significantly worse oral health than the control group, as confirmed by higher values of the D, DMFT, PI, and BOP indices. It was found that the main factor influencing the composition of the oral microbiota was antibiotic therapy, and not the SARS-CoV-2 infection itself. The increased ratę of pro-inflammatory pathogens (Prevotella, Capnocytophaga, Eikenella, Tanerella, Selenomonas) observed in C0VID-19 patients highlights the importance of preventing periodontal disease and improving oral hygiene in the futurę. The correct sequence of procedures during dental examinations and sample collection is crucial to avoid technical errors and contamination ofthe collected materials. The DMFT index, D number, PI, and BOP indices are effective prognostic parameters of oral health, and the procedures developed as part of the project can be used as ready-made forms in futurę studies.
