Research clearly indicates that exposure of the offspring at an early stage of development to an unbalanced maternal diet may lead to diseases m adulthood, including neuropsychiatrie diseases. Since these diseases are often accompanied by cognitive disorders related to the dysfunction of the NMDA receptor complex, it was purposeful to investigate whether NMDA receptor dysfunctions could also occur m offspring prenatally exposed to an unbalanced matema! diet. The studies were performed in a rat model of prenatal and early postnatal exposure to the high-sugar or high-fat diet of the mother. Behavioral tests showed significant disturbances, especially in spatial memory. Molecular studies of the medial prefrontal cortex and hippocampus revealed changes in the expression of NMDA receptor subunits, associated scaffold proteins, and microRNAs regulating NMDA receptor activity, as well as changes in extrasynaptic glutamate levels and the expression of its transporters. The o btained results confirm that the calorically unbalanced diet of the mother may contribute to the occurrence of behavioral disturbances in the offspring as well as changes in the expression and regulation of -~MDA receptor. Demonstrating the key role of the maternal diet in the proper development of the offspring may contribute to increasing public awareness of the role of nutrition dur ; ing pregnancy and lactation.
