Title: The role of cancer-specific isoenzymes of phosphofructokinase II in the progression of malignant melanoma including the role of low-oxygen environment of tumor development

Abstract:

One of the glycolytic enzymes which expression seems to alter in carcinogenesis is phosphofructokinase II (PFK-II). In cancer cells the expression of two cancer specific isoenzymes of PFK-II encoded by PFKFB3 and PFKFB4 genes is commonly observed. The research conducted in the frame of PhD thesis, on the panel of melanoma cell lines, revealed that PFKFB4 protein expression may be one of the main mechanisms modulating glycolytic activity in hypoxic malignant melanoma cells. Additionally, the analyzes revealed that high expression of the P FKFB4 gene is associated with poor prognosis of cancer patients not only in melanoma, but also in several different tumors. The research presented in the thesis also revealed that the expression of the PFKFB4 gene in melanoma cells does not reflect the expression at the level of individual isoforms. PFKFB4 isoform D and F are expressed as the constitutive ones, whereas isoform B seems to be the main one induced in hypoxic conditions. The obtained results suggest the importance of PFKFB4 isoforms' expres si on in metabol ie adaptation of malignant melanoma cells, by directing the glucose flow by two important metabolic pathways: glycolysis or the pentose phosphate pathway and indicate these proteins as promising molecules in targeted anti-cancer therapy.
