CD146 (MCAM), described as a protein that induces epithelial-mesenchymal transition (EMT), is present in an increased amount in cancer cells. So far, however, the cause of the increased expression of his gene has not been elucidated, excluding the involvement of genetics, which suggests that its regulation may be influenced by epigenetics. The research undertaken as part of the work was aimed at explaining the role of methylation in the regulation of CD146 gene expression. CpG island methylation analysis of the promoter region was performed by MS-PCR and BGS in prostate and breast cancer cell lines, revealing heterogeneous methylation in MCF7 and MDA-MB-231 lines. The demethylating compound increased the expression of CD146 in all tested cell lines. Additionally, analysis of clinical data confirmed the relationship of CD146 expression with EMT in both types of cancer. The results of the study allowed for the verification of literature data and confirmed the role of the epigenetic mechanism in the control of CD146 expression. These results are particularly important in the face of the growing importance of epigenetics both in diagnostics and oncological prognosis.
