Title: Perivascular adipose tissue inflammation in regulation of endothelial function in human blood vessels

Abstract:

Most of the arteries of the body are surrounded by perivascular adipose tissue (PVAT), which affect their function. In diseases such as atherosclerosis PVAT reveals features of inflammation, while physiologically acts vasoprotectively. However, a detailed characterization of the cells present in PVAT and mechanisms leading to endothelial dysfunction are not known. The present work investigates a detailed characterization of PVAT T cells in patients undergoing CABG, analyzes relationship between perivascular inflammation and adipose tissue phenotype and its influence on endothelial function. The majority of T lymphocytes in PVAT were TRM cells which do not return to circulation. They predominate especially around atherosclerotic coronary arteries, what is caused mainly by IFNγ producing CD8+ cells. CD4+ TRM cells produce more inflammatory cytokines than cells returning to circulation. Treg lymphocytes in blood, but not in PVAT, are associated with better endothelial function. PVAT surrounding coronary arteries reveals features of inflammation, incomplete differentiation and white adipose tissue phenotype. Inflammatory infiltration to PVAT is associated with increased expression of genes connected with oxidative stress, inflammation and selected adipocytokines. PVAT and IFNγ directly deteriorate endothelial function which depends on perivascular inflammation intensity.
