Polygonatum verticillatum (L.) All. contains over 3% of diosgenin in its underground parts and it is, up to now, the only plant growing in the moderate climatic zone which has such a high content of that aglycone. Diosgenin is used in the semisynthesis of about 80% of steroidal hormones produced m the world. The investigated species may be of use in therapy, especially as a source of semiproducts used in the synthesis of sexual hormones and corticosteroids. The aim of this research work was to study the effect of influence of plant growth regulators, steroidal hormones and cultivation conditions on the induction and growth of callus tissue of Polygonatum verticillatum (L.) All., as well as on the diosgenin content. The experiments were performed on a specially prepared in vitro cultivation of this species. It was found, that the most favourable effect on the callus tissue induction from the hypocotyl parts of plantlets is obtained, when 2 mg/dm3 of 2,4-D and 1 μM/dm3 of 17- β-estradiol are added to the Murashige and Skoog medium (MS). Quantitative analysis of diosgenin was carried out using the TLC-densitometry. The obtained results indicate a higher content of diosgenin in the callus tissue (average 0,1717%) in comparison to the overground parts (shoots in vegetation period - average 0,0122%, ripe fruits - average 0,0219%), but lower content of diosgenin in comparison to rhizo ; mes (in vegetation period average 1,2211 % ). The content of diosgenin in the rhizomes collected in August (1,4040%) was the highest, thus this month seems to be the best for obtaining this plant material.
