Title: Effect of selective 5-HT6 serotonin receptor ligands on the activity of antipsychotic drugs in rats

Abstract:

The aim of this study was to assess the effects of combined administration of a selective 5-HT6 receptor agonist (WAY-181187) or antagonist (SB-742457) with selected antipsychotic drugs (APD), i.e. haloperidol, risperidone, and olanzapine in in vivo and ex vivo studies in rats. Behavioral studies assessed effects of the tested compounds and their combinations on different cognitive functions disturbed by MK-801. In addition, their potential antidepressant and anxiolytic activities were assessed. Ex vivo studies included the assessment of the effect of the tested compounds and their combinations on the level of the brain-derived neurotrophic factor (BDNF) protein and its gene expression in the hippocampus and prefrontal cortex. 5-HT6 receptor ligands modified the activity of APD against memory deficits in various ways, depending on the test used. In addition, WAY-181187 reduced pro-depressive and anxiogenic effects of haloperidol. Both ligands, administered with risperidone, revealed an anxiolytic effect. The addition of 5-HT6 receptor ligands to olanzapine had no significant effect on the depressive and anxiety behavior of animals. The BDNF signaling pathway is not the only mechanism associated with the demonstrated pro-cognitive, antidepressant and/or anxiolytic activity of the tested substances and their combinations.
