Abstrakt:

The aim of the study was to characterize some processes related to hydration of matrix tablets on the example of tablets with sodium alginate. The studies were carried out on model tablets with sodium alginate (ALG) or its mixture with salicylic acid (SA) or sodium salicylate (SNa) (1:1), which were subjected to hydration in water or hydrochloric acid (0.1 mol/dm3) solution for four hours. The distribution of water in tablets - studied by Karl-Fischer method - depended on the type of drug substance included in the tablet and the properties of the incubation solution. SNa increased water penetration into the matrix and SA delayed its hydration. DSC studies showed that non-freezing water in tablets containing ALG or polymer and SA was present in the hydration layer of matrix components. In case of tablets containing SNa it existed also in the pores of their external layer. MRI studies showed that proton mobility and proton density in particular matrix areas depended on the tablet composition. Exploratory analysis of the relationship between the release of drug substances and the distribution of absorbed water in the matrix showed a linear, directly proportional relationship between the release of SNa and the content of water absorbed in the deeper layers of the matrix. In the case of tablets with SA this relationship concerned its external layer and was inversely linear.
