Cow's milk is one of the most common causes of food allergies among children, and caseins constitute 80% of its proteins. There is a group of patients in whom allergic reaction to cow's milk is delayed, possibly cell-mediated. Recent discoveries have shown that excessive cellular response can be inhibited using specific suppressive exosomes with miRNA-150. The aim of the research was to describe the mechanism of delayed-type hypersensitivity (DTH) reaction to casein in mice and to evaluate the potential therapeutic application of casein-specific suppressive exosomes. In the present study, it was shown that DTH reaction to casein in mice is mediated by CD4 + Th1 lymphocytes and macrophages. The suppressive properties of casein-specific exosomes were confirmed in adoptive transfer in vitro and in a mouse model of DTH to casein in vivo. The strongest suppression effect was obtained with the oral route of exosomes administration. doktorat
