Abstrakt:

The aim of this study was to evaluate antipsychotic-, antidepressant- and anxiolytic-like activity in the group of 8 arylpiperazine alkyl derivatives of salicylamide with significant affinity for dopaminergic D2, serotonergic 5-HT1A, 5-HT2A and 5-HT7 as well as adrenergic α1 receptors. Multiple behavioral studies in mice were performed, such as: hyperlocomotion tests induced by MK-801 and amphetamine to evaluate antipsychotic-like activity, forced swimming and tail suspension test to determine antidepressant-like properties as well as four plate and the elevated plus-maze test to verify anxiolytic-like activity. Additionally, the influence on spontaneous locomotor activity, motor coordination and cataleptogenic activity in mice were verified. Moreover, the effect on cardiovascular system was investigated. Based on the obtained results, two most active derivatives were selected for extended studies, such as: open field and the prepulse inhibition test after MK-801 administration in rats to determine antipsychotic-like activity, marble burying test in mice to evaluate anxiolytic-like properties as well as passive avoidance and novel object recognition test to investigate the influence on cognitive function. The results of the behavioral studies confirm potential antipsychotic-, anxiolytic- and/or antidepressant-like activity as well as possible procognitive effects of the teste ; d compounds in animals. This encourages to continue studies with selected derivatives, in order to understand their full pharmacological and safety profile.
