Title: Detailed characteristic of stem cells isolated from Wharton's Jelly (WJ-MSCs) by use of modern molecular genetic methods

Abstract:

The subject of the study were stem cells isolated from Wharton jelly and assessment of their proangiogenic properties in the treatment of hindlimb ischemia. The study showed that WJ-MSCs meet the criteria set by ISCT: a typical MSCs cell phenotype and the potential for proliferation and differentiation. In addition, they express the numerous adhesion molecules, pluripotent factors and therapeutically important proangiogenic factors. The obtained results confirm the high regenerative potential of WJ-MSCs. The second part of the work concerned the assessment of the regenerative potential of WJ-MSCs in the in vivo model. An effective model of hindlimb ischemia was developed in NOD-SCID strain mice by ligation of the femoral artery. The cells ability to engraftment the injury site was examined using in vivo bioluminescent imaging. The experiments performed showed improvement in WJ-MSCs-treated mice compared to controls. The positive effect was confirmed by behavioral observations, as well as an increase in the level of blood flow measured by the Laser Speckle method. Histological analysis also showed regeneration of muscle tissue after WJ-MSCs transplantation. Administration of WJ-MSCs cells has proved to be a safe method that can be used to treat civilization diseases, especially cardiovascular diseases.
