Radiotherapy is one of the leading and dynamically developing cancer treatment modalities. Its main aim is to kill cancer cells while sparing normal tissue. The goal of the study was to assess the usefulness of cytogenetics and dose calculation algorithms in predicting radiotherapy effectiveness. Human peripheral blood lymphocytes were irradiated ex vivo in the dose range of 0.3-20.0 Gy. The analysis of cell radiosensitivity was performed using 2 cytogenetic tests: micronucleus (CBMN) and premature chromosome condensation (PCC). Also, the compliance of the results obtained by dose calculation algorithms with those actually measured or given by the Monte Carlo simulation was evaluated.Scoring of 500 binucleated cells instead of 1000 for the CBMN test and 150 instead of 75 for the PCC test showed statistical compatibility, which shortens analysis time. Comparing the 2 dose calculation algorithms: PBC and AAA with the values measured in structures of high electron density gradient, shown that the use of the AAA algorithm gives convergent results with the values measured and obtained from the Monte Carlo simulation.The use of cytogenetics and appropriate dose calculation algorithms will help optimize and predict the effectiveness of radiotherapy.
