The research was funded by a grant from the National Science Centre (2012/05/N/NZS/00846 to Piotr Mazur). The priority was to evaluate the role of coagulation, fibrinolysis impairment, immune system involvement and genetic regulation of valvular calcification in a cohort of patients being surgical candidates. Because valvular heart disease is associated with increased thromboembolic risk, and impaired fibrinolysis was reported in severe AS, while data about fibrinolysis impairment in degenerative mitral stenosis (MS) were scarce, we first sought to compare fibrinolysis impairment in AS and MS. Then, we sought to investigate the role of different circulating lymphocyte and monocyte subpopulations, with focus on CD4+CD8+ lymphocytes and natural killer T (NKT) cells, in AS. Finally, because valve calcification can be excellently evaluated by ex-vivo microcomputed tomography (micro-CT), we decided to investigate the associations between micro-CT findings and biological indices of calcification in severe AS.
