Gastritis can be induced by many factors of both endogenous and exogenous origin. The pathophysiology of this process is related to the presence of reactive oxygen species (RFT). The main factors contributing to the development of inflammatory changes in the gastric mucosa which lead to the disturbance of a delicate oxidative-anti-oxidative balance are: the reduced level of antioxidant defense involving both enzymatic and non-enzymatic mechanisms, ingestion of exogenous irritants, stress, bacterial infections and vascular disturbances related to ischemia-reperfusion phenomena (I/R). The aim of the doctoral thesis was to determine the effect of rich or L-tryptophan (L-Trp) free diet on the healing process of the experimental acetic ulcer in rats. This research attempted to study whether the abundance or lack of L-Trp, which is considered as the precursor of melatonin (MLT) biosynthetic pathway, can influence the formation and gastric tissue levels of endogenous MLT. Moreover, the biochemical pathways of melatonin, the indoleamine with a potent anti-inflammatory and antioxidative protective potentials was investigated. For this purpose, rat gastric mucosal biopsies were taken from the inflammatory sites at ulcer margin in the phase of ulcer healing. The obtained results prove that the rich L-Trp diet accelerates the process of gastric ulcer healing and inhibits the peroxidation o ; f PUFA possibly due to the gastrointestinal conversion of L-Trp to anti-inflammatory and antioxidazing melatonin.
