Title: Development of a panel of protein biomarkers and their application to multi-parameter assessment of vascular endothelial function

Abstract:

Healthy endothelium is essential for the proper functioning of the cardiovascular system, and its dysfunction is the cause or the consequence of many diseases. The aim of the study was to develop and validate the method for simultaneous determination of a panel of endothelial specific protein biomarkers using targeted proteomics. Furthermore, the goal was to assess the changes in biomarker concentrations in the diseases with confirmed endothelial dysfunction in mouse models of fatty liver caused by high-fat diet, endotoxemia induced by intraperitoneal administration of lipopolysaccharide and metastatic breast cancer. The developed methodology was also used to determine the plasma biomarker concentrations of healthy volunteers, patients with chronic myeloid leukemia treated with imatinib and patients with obstructive sleep apnea. The developed panel included markers involved in processes specific for glycocalyx disruption, endothelial inflammation, increased vascular permeability, dysregulation of hemostasis and other processes related to endothelial dysfunction. The elaborated targeted proteomic method for determination of selected mediators released by the vascular endothelium has the potential to be used in humans as a new tool in the early diagnosis of diseases with confirmed vascular endothelial dysfunction and to monitor the effectiveness of the therapy.
