Title: Prognostic significance of mean platelet volume and its genetic variants in Polish patients with ischemic stroke


Increased mean platelet volume (MPV) is a marker of worse outcome in cardiovascular disorders. The aim of the study was to determine whether MPV and single nucleotide polymorphisms (SNPs) associated with MPV in genome-wide association studies (GWAS) can be prognostic markers in patients with acute ischemic stroke. Demographic, clinical and laboratory data was collected in 577 patients with first-ever ischemic stroke. Genotyping of three SNPs (rs342293, rs1354034 and rs7961894) was performed in all subjects using a real-time polymerase chain reaction allelic discrimination TaqMan 7900 assay. Among three tested SNPs only rs7961894 was independently associated with MPV in patients with acute ischemic stroke and allele T of rs7961894 remained an independent marker of a lower one-year mortality after ischemic stroke (HR=0,27; 95%CI:0,09-0,77; p=0,014). MPV was not associated with one-year mortality in patients with ischemic stroke, however MPV ≥11,6fl was independently associated with a worse stroke severity based on National Institutes of Health Stroke Scale (NIHSS) score on admission in the studied population (OR=1,68; 95%CI:1,05-2,68; p=0,031). MPV and rs7961894 were not associated with functional independence assessed by modified Rankin Scale or activity of daily living assessed by Barthel Index on discharge in the studied population.
