The primary aim of the presented paper is to determine the influence of testosterone on the opioid system activity in lean polycystic ovary syndrome (PCOS) women. The β- endorphin plasma levels as well as the pressure pain thresholds (PPT) in lean PCOS women and controls were measured and the associations between these parameters were analyzed. Furthermore the influence of metformin therapy on β- endorphin plasma levels and PPT values in lean PCOS women and controls was investigated. Higher β- endorphins level and correlations between β- endorphins concentration and other hormonal parameters in PCOS group were revealed. We also observed higher PPT values measured on deltoid and trapezius muscle in PCOS group, moreover PPT values were positively correlated with β- endorphins plasma levels. Furthermore an increase in PPT values after 6 months metformin administration in PCOS group was observed. These results can indicate the β- endorphins role in PCOS pathophysiology, and also suggest that the increases in circulating plasma β- endorphins concentration can increases PPT values in PCOS women group. Moreover we conclude that metformin administration increases PPT values in PCOS lean women. It maysuggest the potential role of metformin in pain perception in humans that was reported in several earlier findings.
