Title: Gastroprotective activity of carbon monoxide in experimental models of gastric mucosa injury

Abstract:

Carbon monoxide (CO) is a gaseous mediator with pleiotropic biological activity. CO is produced intracellularly during heme degradation in reaction catalyzed by heme oxygenase (HO). CO has been considered to possess neuromodulatory, vasodilatory, anti-inflammatory and anti-aggregatory properties. Growing body of evidence points out to the important role of CO as a component of complex mechanism of gastric mucosal defense. The major aim of my doctoral dissertation consisting of three papers published in international journals was to determine the role and physiological significance of CO and HO-1 in pathomechanism of acute gastric lesions formation induced by 75% ethanol or water immersion and restraint stress (WRS). In brief, the present study has demonstrated that CO generated endogenously or that released from its chemical donor (CORM-2), contributes to gastric mucosal protection against local and systemic damage caused by ethanol and stress, respectively. Hypermic effect of CO involves the activation of sGC/cGMP system, biosynthesis of endogenous PGs by COX-1 and COX-2 and, at least to some extent, NOS/NO system and CGRP-releasing afferent sensory nerves activity. The enhanced bioavailability of CO prevents lipid peroxidation and limits inflammatory response exerting anti-oxidative activity in gastric mucosa subjected to damaging factors.
