Title: Induction of apoptosis in melanoma cells influenced by inhabitors of the inner signalling pathways

Abstract:

Melanoma is characterized by a high mortality rate and resistance to commonly used treatment. Intensive research regarding melanoma are conducted in targeting of signal transduction pathways and its molecules that regulates apoptosis. Many of the research are recently focused on the inhibition of heat shock proteins family. Numerous studies indicate that many of the therapeutics commonly used in cancer treatment, achieve their therapeutic effect by induction of apoptosis. In addition, it is strengthened by use of different proapoptotic factors.. The study investigated the ability of inhibitors of signaling pathways combinations and inhibitors of heat shock protein Hsp90 to stimulate apoptosis in melanoma cells. Studies were conducted on melanoma cell lines from different stages of tumor progression. The number of cells was determined by crystal violet and trypan blue staining, the disintegrity of cell membrane by the LDH cytotoxicity test, the process of apoptosis by caspase-3 fluorimetric assay and the TUNEL test, the level of the activated caspase-9 was determined by Western Blot. Particularly strong proapoptotic effect was observed for the combination: 100μM olomucine + 20µM cytochalasin D + 50μM LY294002 and 50nM 17AAG + 2.5µM R115777. This suggests a synergistic effect of the combination of inhibitors for induction of apoptosis. The selected combination tested in further c ; linical trials, may become a potential candidate for the treatment of melanoma.
