Tytuł: The influence of HGF/MET axis on EMT activation in metastatic progression of cervical cancer
Abstrakt:
Survival of women suffering from cervical cancer is directly negatively connected with metastatic potential of tumor cells. The activation MET receptor via hepatocyte growth factor HGF is responsible for epithelial to mesenchymal transition (EMT) in numerous tumors. Morphological changes occurring during EMT lead to acquiring malignant and invasive abilities of expanding tumor.The aim of this dissertation was to evaluate the influence of HGF/MET axis activation on metastatic progression of different stages of cervical cancer. Determination of signaling pathways responsible for this effect may indicate potential therapeutic targets.We used C-4I and HTB-35 cervical cancer cell lines, which biological properties correspond with different tumor stages. We examined intracellular pathways responsible for cells scattering byblocking signal transduction cascades. Gene expression patterns related to metastatic progression were evaluated by Real-Time PCR analysis and Western Blot. Differences in cellular localization of adhesion molecules and cells cytoskeleton organization were estimated by immunofluorescent staining, while morphological changes were examined with light microscope and holographic microscopy.Summarizing, HGF/MET axis is fully responsible for acquiring malignant traits on cervical cancer cells, at both early and late stages of the disease. Recognition of molecular mechani ; sms responsible for tumor metastasis may be useful in improving treatment efficiency.
