Abstrakt:

This dissertation presents the effect of statins, angiotensin converting enzyme inhibitors (ACEIs) and angiotensin II receptor blocker (ARB) on the expression of proteins associated with inflammation, calcification and coagulation cascade in vitro in VICs cultures. This study presents mRNA analysis of the gene expression associated with inflammation, calcification and osteoblastic transformation, coagulation proteins, PAR receptors and a neovascularization using Real-time PCR in three different aortic valve areas. In addition, the influence of statins: atorvastatin and rosuvastatin, ACEI: enalpril and ramipril, and ARB: candesartan, in different concentrations on the expression of proinflammatory cytokines, calcification and expression of coagulation proteins and PAR receptors in stimulated VICs was analyzed. This in vitro study increases knowledge about the effect of statins, ACEIs and ARB on proinflammatory response, calcification and expression of coagulation protein in AS.
