Title: Influence of new biocompatible, cationic polysaccharides on lipid metabolism in laboratory animals - efforts of determination of action mechanism

Abstract:

The aim of this PhD project was to test the newly synthetized, modified cationic polysaccharides on their antiatherosclerotic properties and their effects on lipid metabolism. Efforts have been also attempted to explain their mechanism of action.The physicochemical and biological properties of the cationic polysaccharides were examined in experiments in vivo and in vitro. It has been shown that cationic polysaccharides such as HPC-APTMAC, Pull-GTMAC, HTCC and Dex-GTMAC are biologically active.Cationically modified polysaccharides administered orally to apoE-knockout mice (murine model ofatherosclerosis) at a dose of 300 mg/kg b.w./day for 18 weeks (HTCC and Pull-GTMAC) have anti-atherosclerotic potential, reducing the area of atherosclerotic plaque and affecting lipid metabolism. HTCC at a dose of 200 mg/kg b. w./day administered to mice for 16 weeks reduces the level of cholesterol in the plasma. HTCC and other cationic polysaccharides affect the expression of lipid metabolism genes (HMG-CoAR and LDLR), both in vitro on cell lines (HepG2 and RSM) and in studies on the material from the apoE-knockout mice in vivo.
