Tytuł: Application of human induced Pluripotent Stem Cells in therapy of neurodegenerative diseases based on example of animal model of Parkinson’s disease

Abstrakt:

Parkinson’s disease (PD) is the second most frequent neurodegenerative disorder. Present therapeutic strategies are symptomatic and their effectiveness decrease in time. Among most promising approaches which could not only improve the symptoms but also cure the cause of the disease regenerative therapy involving induced pluripotent stem cells (iPS) appears to be most encouraging. Main goal of the study was to assess the usefulness of iPS cells in PD therapy. iPS cells specific for healthy volunteers and PD patients were acquired and characterized. Next, iPS cells were differentiated into dopaminergic neurons. Also, cells producing black pigment – melanin were acquired. Thus, a model enabling deciphering pathomechanisms of PD was generated. Subsequently, differentiated dopaminergic neurons were grafted into the brain of rat model of PD. Promising integration of grafted cells with surrounding brain tissue was observed. Murine model of PD with severe immunodeficiency was also established. In the last part genetic safeguard allowing specific elimination of all grafted cells in case of their malfunction was developed. Summarizing, in this study we present that regenerative therapy of PD involving iPS is very promising strategy and may become an alternative for present therapies of neurodegenerative diseases.
