Abstrakt:

Despite of good understanding of mobilization of hematopoietic stem cells it is still difficult to yield sufficient number of peripheral blood stem cells for transplantation in some patients. The objective was to assess the influence of G-CSF dose on mobilization efficacy in experimental model in different mice strains and in clinical observation in children and to find out other factors influencing the mobilization efficacy. Four mice strains were stimulated with G-CSF in different doses. White blood count, number of hematopoietic cells and clonogenity were assessed and syngenic transplantations were done. Retrospective analysis was performed in 82 patients. Collection of a minimal and optimal CD34+ cells count were assessed. There was observed that an optimal dose of G-CSF for each strai exists Different patterns of mobilization between different mice strains were observed. In the clinical analysis no influence on the mobilization efficacy of G-CSF dose, way of mobilization, type of disease and time from diagnosis to mobilization was observed. The mobilization was more efficient in boys. In multivariate analysis the influence on the mobilization efficacy of G-CSF dose was not observed. The time from diagnosis to mobilization and a type of disease were statistically significant factors and influence of sex was observed as a tendency. The mobilization was ineffective in 15%.
