Title: Induced pluripotent stem cells derived from somatic cells and their application in treatment of osteogenesis imperfecta

Abstract:

In recent years considerable attention has been put on pluripotent stem cell research. The research has been facilitated even more by a publication in which a research group lead by Shinya Yamanaka presented possibility of reprogramming somatic cells into pluripotent state by ectopic gene expression. The generation of induced pluripotent stem cells has given new hopes on establishing convenient and potent source of cells for correction of genetic disorders and regenerative medicine. The thesis presents results of attempts to generate induced pluripotent stem cells with retroviral and lentiviral vectors. Mouse embryonic fibroblast isolation, subculturing and feeder layer preparation is also described. An outcome of non-invasive method of obtaining cell material from human donors is also depicted. After that, unsuccessful attempts of somatic cell reprogramming with lentiviral vectors are presented and discussed. Results of reprogramming of human fibroblasts and keratinocytes with retroviral vectors are also presented. Furthermore, a comparison of viral vector suitability for reprogramming of somatic cells is shown. Finally, a perspective of harnessing the obtained pluripotent stem cells in creating an in vitro gene therapy model for inborn brittle bone disease – osteogenesis imperfecta is discussed.
