Tytuł: Evaluation of the hemotoxic and gonadotoxic effects of selected alkoxyethanols in rats

Abstrakt:

Alkoxyethanols, such as methoxyethanol (ME), ethoxyethanol (EE), and butoxyethanol (BE) are widely used as mixtures in industry and households. The objective of the present study was to evaluate the hematological effects in peripheral blood, and also gonadotoxic actions in rats treated with the mixtures: ME and EE, BE and ME, and BE and EE at low dose levels with molar ratio 1:1 for 28 days.A short-term repeated exposure of rat to ME and EE alone led to weak hematological alterations increasing with exposure time. In the rats co-exposed to ME and EE additivity effects were observed. BE alone induced hemolysis; hematological alterations were most pronounced at the beginning of the exposure, later these changes clearly regressed despite continued exposure to this ether. In the rats co-exposed to BE and ME or BE and EE significantly lower hematological changes were observed at the beginning of the exposure in comparison with animals treated with BE alone. In the later period, i.e., at the end of a 4-week exposure, the hematological alterations in the same animals were markedly pronounced and progressively elevated with time of drug exposure. The relative weight of testes, epididymes, and prostate negatively correlated with doses of ME (a non-linear regression). Similar effects were observed in relation to a decrease in protein, and tiols levels in testis. EE alone exerted weak gon ; adotoxic effects. Co-exposure to ME and EE let to independent gonadotoxic effects. The mixture of ME and EE did not affect the glutathione-dependent antioxidant enzyme activity, total antioxidative status, and malon-dialdehyde level. Neither ethers nor their mixtures induced lipid peroxidation in testes. In the paper the possible mechanisms of observed phenomena were discussed.
