Title: Clinical value of sHLA-G designation in pregnant women

Abstract:

OBJECTIVES: The objective of dissertation is to explain changes of sHLA – G concentration in pregnant woman’s serum during labor and their connection with activity of mother’s immune system.MATERIAL AND METHODS: 177 patients were qualified for examination. In the examined groups were included patients from which peripheral blood for examination in cases in which changes in the activity of pregnant mother’s immune system can be observed ( labor, premature labor, spontaneous miscarriage, ectopic pregnancy ). For detecting sHLA-G In blond serum was applied examination set using ELISA technique. In addition, characteristic of lymphocytes T CD57+ and NK CD56+ cells population in decidua was made with use flow cytometry examination.RESULTS: The presence of sHLA – G was indicated at all patients being in the beginning of labor or in active phase of labor. In initial phase of labor was observed statistically indicative increase of sHLA – G concentration, whereas in final phase of labor was observed statistically indicative decrease of sHLA – G concentration in blood serum. In addition, it was indicated that beginning of labor means the increase of population of CD56+ and CD57+ cells in deciduas, and in progress of labor was observed decrease of those subpopulations NK cells and T lymphocytes along with increase of HLA – G antigen in blood serum. Similarly, in first trimester of pregnan ; cy while development of ectopic pregnancy was observed statistically indicative increase of sHLA – G in blood serum. Its level was comparable to the one observed during spontaneous miscarriage, whereas during rupture of the side of the Fallopian tube concentration of sHLA- G in blood serum statistically indicatively decreased.CONCLUSIONS: The concentration of HLA – G antigen can reflect increase ofphenomenon of selective suppression of mononuclear cytotoxic cells of the immune system which are in decidua’s microenvironment. Measurement of this antigen’s concentration can be used clinically as potential sign of changes in microenvironment of decidua and endometrium.
