Abstract:

The study was undertaken to evaluate the metabolic and toxicokinetic interactions between two-component mixtures of 1,1,2,2-tetrachloroethylene, ethylbenzene and n-hexane administered subcutaneously (28 days) or with drinking water (3 months) to Wistar rats. The scope of the study included: tested compound metabolism evaluation (the concentrations of 2.5-hexanedione, hippuric acid and trichloroacetic acid in daily urine samples were measured), neurotoxicity assessment in behavioral tests (corneal and righting reflex, psychomotor performance in open field, functional condition of the mobility organ in the rotating rod test, antinociceptive activity in a hotplate test and exercise test on treadmill) and hepatotoxicity determination with biochemical indicators (ALT, SDH, G1DH, GGTP) and serum lipid fractions. MD A and NPSH were measured to evaluate prooxidation mechanisms. The results of the 28-day experiment proved metabolic interactions between the compounds tested. In the other hand, hepatocellular damage may occur after 3 months of tetrachloroetylene administration. Most prominent, both in the repeated short-term and subchronic experiment were organ changes manifested by hepatic and renal NPSH decrease. The changes were associated with MDA increase which was indicative of the stimulating effect the tested compounds had on prooxidation mechanisms. It can be said that the key el ; ement in evoking metabolic interactions between the tested compounds is the size of the xenobiotic doses. On the other hand, also the exposure time plays a critical role in the manifestation of pathologic organ changes.
