Tytuł: Influence of genetic polymorphisms of angiotensinogen, angiotensinconverting enzyme and type 1 angiotensin II receptor on large artery stiffness parameters

Abstrakt:

The aim of this study was to establish the frequency of alleles and genotypes of the G-6A AGT, D/1 ACE, and A1166C AT1R gene polymorphisms in the population of the Cracow region families and in subjects with normal blood pressure values and with hypertension. Moreover, an attempt, based on association and family-based analysis, has been made to detect association of the analyzed polymorphisms with blood pressure and with parameters of large artery wall stiffness, as well as to assess the interaction between genetic and environmental factors and gender and their joint influence on the parameters of blood pressure and arterial remodeling. The frequency of alleles and genotypes of G-6A AGT, O/l ACE and A1166C AT1R gene polymorphisms, in the population of families as well as in the subjects with normal blood pressure and with hypertension, is similar to the observed in other European populations. In the examined group, D/I polymorphism of the ACE gene as opposed to G-6A AGT and A1166C AT1R gene polymorphisms, has revealed significant association with selected parameters of blood pressure and arterial remodeling, with the negative effect of the I allele. In relation to the parameters of blood pressure and arterial wall stiffness, the interactions between D/I polymorphism of the ACE gene and daily sodium excretion in the urine were revealed, which indicated that D allele has a prot ; ective role in the group of subjects with high daily sodium intake. In the sample of the Cracow region population, also the interactions between D/I polymorphism of the ACE gene and A1166C polymorphism of the AT1R gene reached statistical significance for systolic and pulse pressures, revealing joint negative effect of I and C alleles.
