Tytuł: Studies on mechanisms of protective action of probiotics from species Lactobacillus rhamnosus on the course of acute diarrhoeas in children

Abstrakt:

Acute diarrhoeas in children, especially until the 2nd year of age, are an important problem globally, including Poland. Studies on the effectiveness of action of probiotic bacteria (mainly about Lactobacillus rhamnosus GG) in various medical conditions of the alimentary canal are carried out widely throughout the world. The aims of the undertaken studies were to: examine other strains from L. rhamnosus species; estimate their in vivo probiotic action, based on the effective colonization of the G.I. tract as well as on the elimination of pathogenic factors; explore the examined strains’ interaction with mucin (cellular mucus) and their influence on the secretion of pro- and anti-inflammatory cytokines. The examined probiotic strains from Lactobacillus rhamnosus species are effective in the treatment of acute diarrhoeas with rotaviral etiology and they also shorten the course of diarrhoea by over 24 hours in the age group: from the 2nd month of life until the 6th year of life. The strains from the administered mixture colonize the digestive tract in 80% of patients during the course of acute diarrhoea and in 40% of patients the colonization is present 14 days after the first dose. The administered L.rhamnosus strains have an influence on the formation of intestinal flora in children. It has been shown, that molecular methods of analysis (PFGE, FISH) are useful in estimation of c ; olonization of the alimentary canal since they help to differentiate the given Lactobacillus strains from the patients’ own flora. In vitro studies confirmed that the examined L.rhamnosus strains show strong antagonistic properties against bacteria pathogenic to the G.I. tract as well as adhesive properties towards the cells of the intestinal epithelium. It has also been shown that a characteristic of microorganisms of the faecal flora is the ability to degrade mucin (cellular mucus) which is not the property displayed by the examined probiotic bacteria. L.rhamnosus strains also have a protective effect by increasing the level of synthesis of antiinflammatory cytokines.
