Title: Gastric epithelial cells apoptosis in environment of Helicobacter pylori


Helicobacter pylori (Hp) is a spiral Gram-negative bacterium that colonizes the human gastric mucosa. Heat shock proteins (HSPs) are crucial for the maintenance of cell integrity under normal cell growth and at pathophysiological conditions such as colonization of gastric mucosa by Hp. This study was designed to determine the expression for HSP70, pro-apoptotic Bax and anti-apoptotic Bcl-2 induced by two live strains of Hp (cagA+ vacA+) and Hp (cagA- vacA-) with and without incubation with exogenous CagA in the two human epithelial cell lines MKN7 and KATO III. In addition, the expression of COX-2 mRNA in MKN7 cells infected with Hp and the effects of COX-2 suppression by the selective COX-2 inhibitor, NS-398, on cell apoptosis were examined. It was found that Hp inhibits expression for HSP70 in MKN7 and KATO III cells but the epithelial cell response may depend on the genotype of Hp strain. When the MKN7 and KATO III cells were co-incubated with Hp and exogenous CagA, the inhibition of HSP70 was much more pronounced as compared with that without CagA addition. Hp-infected epithelial cells without or with CagA showed the upregulation of Bax mRNA and protein and a simultaneous fall in the expression for Bcl-2 at the level of mRNA and protein. Hp strain (cagA+ vacA+) upregulated COX-2 mRNA and the expression of COX-2 and Bax mRNAs was inhibited by NS-398. This study indicates tha ; t Hp stimulates apoptosis via mechanism involving inhibition of HSP70 and activation of COX-2.
