Abstract:

The main aim of present studies was to define the in vitro sensitivity ofHL60 and NB4 cell lines towards arabinoside cytosine, daunorubicine, mitoxantrone and etoposide and investigate whether differentiation of these cells induced by A TRA and PMA alters their drug sensitivity. The drug effect was examined by cytofluorimetric measurement using Alamar Blue assay (determination of proliferation and survival rates) and evaluation of cell smears stained with Hoechst 33342 (calculation of apoptosis index). In the first part of studies it was found that: - both cell lines revealed varying sensitivity towards all drugs (cytostatic and cytotoxic effects) - the decrease of cell survival resulted from an induction of apoptosis The differentiation along the neutrophil lineage (upon stimulation with ATRA) and along the monocyte/macrophage lineage (upon stimulation with PMA) was proved by decreased proliferative potential of cells, morphological changes, increased ability for NBT reduction and increased expression of CD lib and CD14. Investigation of chemosensitivity of cells stimulated to differentiation showed that: - ATRA-induced differentiation increased cell sensitivity to arabinoside cytosine and mitoxantrone but decreased their sensitivity to etoposide - PMA-induced differentiation resulted in the decreased cell sensitivity to all drugs tested.
