TY - GEN A1 - Bulanda, Małgorzata N2 - The topic of researches is the application of basophil activation test (BAT) in the qualification and monitoring of specific immunotherapy (SIT) with inhalant allergens.BAT was compared to the results of skin prick test (SPT), specific IgE (sIgE) and nasal provocation test (NPT) in 30 patients with the diagnosis of allergic rhinitis (AR). Basophils were stimulated with allergens in 5000, 500 and 50 SBU/ml concentrations used in NPT.During the SIT, BAT was carried out in 31 children with AR and/or asthma after patients had been administered the same cumulative dose of allergen (BAT2 and BAT3; BAT 1 during the qualifications). Basophils were stimulated with allergens in concentrations from 22.5 ng/ml to 0.00225 ng/ml. BAT results were expressed as a percentage of activated basophils, stimulation index (SI), basophil reactivity (BR), basophil sensitivity (CD-sens).BAT sensitivity, specificity, the positive and negative predictive value in appropriate concentrations in relation to NPT ranged 75-100%. BAT was conclusive in 2 of 3 patients with incompatible results of SPT and sIgE. CD-sens index decreased significantly in all patients in BAT2 and in BAT3 was observed further decrease in this parameter.BAT may be considered as an alternative method in the qualification for SIT. Both SI and BR are credible basophil activation indicators. The CD-sens index is a better monitoring paramet N2 - er than the plain percentage of CD63-expressing basophils. Allergen suitable for NPT in appropriate concentrations is a good reagent for use in BAT.
