One of the prognostic and predictive factors in invasive breast carcinomas is determination of HER2/neu protooncogene amplification or HER2 protein overexpression. Overexpression of this receptor occurs in about 10 to 34% invasive breast carcinomas which is correlated with 90% of HER2 gene amplification. It is associated with more aggressive disease, greater likelihood of recurrence and generally poor prognosis. AIM OF THE STUDY: Sensitivity, specificity, simplicity of a given procedure and method standardization, simplicity of evaluation results of each in situ hybridization method, time needed for performing test and the cost per one slide were compared. MATERIAŁ AND METHODS: 63 cases of infiltrating breast carcinoma from surgically excised tumours and core needle biopsies, embedded in paraffin blocks were included in the study. First step was the determination ofHER2 status by immunohistochemistry (IHC) - HercepTest. The patients with moderate (2+) and strong overexpression (3+) of HER2 protein, were chosen to check HER2 amplification by 3 methods of in situ hybridization: FISH, CISH and in situ hybridization in connection with silver autometalography. RESULTS: The overall concordance rate between IHC and ISH methods was not so high, but statistical analysis revealed a good agreement b}, abstract={etween IHC and ISH methods and among ISH methods (p << 0,01).
