The aim of this study was to investigate antioxidant, anti-platelet and vasoprotective properties of polyphenols from A. melanocarpa. We studied the effects of A. melanocarpa extract on superoxide(O2-.) production in arteries and veins from patients with atherosclerosis, direct superoxide scavenging properties, effects on vascular wall tension, and the effects on platelet functions. We observed that A. melanocarpa extract has direct ROS scavenging effects at high concentrations, while at lower concentrations also decreases baseline production of O2-. in arteries and veins, significantly inhibits NADPH oxidase activity in part in NO dependent manner. A. melanocarpa extracts cause vasorelaxation and the degree of this effect seems to be correlated with NO-dependent endothelial function. Vasorelaxant effect depends on endothelium and seems to be mediated by an enhancement of the NO-cGMP pathway and partially depends on SOD activity. We observed that baseline superoxide production was greater in platelets from patients with atherosclerosis than in controls and that Aronia melanocarpa extract decreased baseline O2-. in resting platelets in this group as well as inhibited platelet aggregation .
