Search for: [Abstract = "The aim of this study was to investigate several adaptive changes after repeated treatment with antagonists of group I metabotropic glutamate receptors\: MPEP and MTEP and to compare these changes with effects induced by conventional antidepressants. Receptor adaptive changes, after chronic administration of MTEP, were determined in rats brain. The results of our studies showed significant α1\-up\-regulation and no influence on β\- and α2\-adrenergic receptor. In glutamatergic system, chronic MTEP administration, produced a significant increase of mGluR5 receptor density in hippocampus and cortex. We also observed reduction of glycine affinity to glycine site of the NMDA receptor complex. Multiple MTEP administration, has no effect on 5\-HT1A and 5\- HT2A serotonergic receptors. Changes in NMDA, α1 and mGluR5 receptors are similar to these induced by antidepressant drugs. Chronic treatment with MPEP, but not MTEP, produced a significant increace BDNF expression in the rat hippocampus. Antidepressant\-like activity of MTEP, in Porsolt test is dependent on 5\-HT2A and NMDA receptors activity. Involvement of 5\-HT1A and AMPA receptors was no observed. The results indicate that interaction between mGluR I and NMDA receptors, as well as between mGluR I and 5\-HT\-receptors play a role in antidepressant mechanism. In conclusion, these data show some similarities in mechanism of action between group I mGluR antagonists \(MTEP\) and conventional antidepressants and suggest their potent activity in clinical depression."]
